When I try to boot my computer a black screen appears with the following
message:

The following file is missing or corrupt: "Windows\System32\Config\System"

Will not go any further and will not open in safe mode either.

Is it a case of reformat the HD.

This is caused by a corrupt system hive. Boot the machine into recovery
console and rename the system hive in the windows\system32\config directory
and use on from backup or the windows\repair directory to reboot the
machine.
