Hello,
is it possible to prevent the access to a folder with a password, as we
can protect a file ?

Only with a third-party tool, but you can deny access to the folder to every
other user except yourself.

You password your profile.
Also you need to be using NTFS and not FAT32 file system.
To check:
Open My Computer, Right click C drive, click Properties.
What does it say by "File System"?
To convert to NTFS:

Data should be safe, back-up important data just in case.
