I have huge files in the folder \WINDOWS\TEMP like a 367
MB file named ~PDT4315.tmp and a 246 MB file named
~PST1163.tmp
Can files like these be deleted without creating a
problem?

Yes, do it after a clean boot, preferably into Safe Mode.

Booting isn't necessary, and certainly not Safe Mode. Deleting
temporary files is always safe unless they are in use (or if
you've just rebooted in the middle of a software installation).
And since Windows won't let you delete an open file, it's safe to
try to delete those files, as well as all the others in the \temp
folder.
