Default Secure folder (process allow, process deny)

I was on the phone with a security tech and he mentioned that it is
possible to secure a folder (set permissions) by restricting the
processes that could access it. like grant an application full control
and deny explore.exe. Is this possible?

I asked him again to make sure I heard correctly but i have not yet
verified this to be true.
